Will Grayson (name always capitalized) begins the novel newly cut off from his Group of Friends, leaving him with no one but his recently acquired best friend Tiny, who is described as being both "the world's largest person who is really, really gay" and "the world's gayest person who is really, really large". Will's group of friends later expands to include will grayson and Jane. Throughout the novel, Tiny is involved with writing, producing, and starring in his autobiographical musical, to be performed at the school. The production of said musical leads to considerable tension between the two.

Much like Will Grayson, will grayson (name deliberately left uncaptalized throughout the novel) is not part of a thriving social circle. However, unlike Will, this is not a new situation for him. His two closest (and only) friends consist of Maura, who goes to his school and is attracted to him, and Isaac, whom he has only interacted with through the medium of the internet.

When will grayson and Isaac decide to meet, an unexpected turn of events brings will grayson, Will Grayson, Tiny Cooper and Jane Turner together. Over the course of the novel, the lives of this unlikely group of people become increasingly intertwined, and everyone involved is changed in ways they would never have imagined.

The odd-numbered chapters (that follow the life of Will Grayson) are written by John Green, and the even-numbered chapters (following the life of will grayson) are written by David Levithan.
