In an early Vlogbrothers video, John had a very important phone call. "So important that he couldn't tell [us] specifically who he was talking to." At the end of the call, instead of saying goodbye, he said "Best Wishes." Then it caught on. He now says it at the end of many videos.
John once said in a HankGames video that in addition to being a funny joke, "Best Wishes" is an interesting salutation because it is another example in the English language of a term having a completely different meaning than one would surmise after reading the words in it. Interestingly, he didn't elaborate, possibly because of an exciting development in a Swindon Town or AFC Wimbledon game.
